Transition Metal‐Free Cycloisomerization of Propargylic Amides to Oxazoles in Hexafluoroisopropanol (HFIP)

چکیده

A transition metal-free method for the cycloisomerization of propargylic amides to oxazoles was developed. The reaction utilizes in situ generated hydrogen chloride hexafluoroisopropanol (HFIP). With aid Design Experiments optimization a wide range substrates transformed into desired oxazoles. allows product formation without side reactions eliminating need extensive work-up and purification.

The synthesis of substituted dihydrooxazoles by the CuI-catalyzed cycloisomerization of terminal propargyl amides is reported. The reaction has been shown to have good substrate scope and experiments to delineate the mechanism have been performed. gem-Difluoro-1,7-enyne amides are suitable building blocks for the synthesis of difluorodihydropyridinones via a ring-closing metathesis reaction, and of 4,4-difluoro-3-oxoisoquinolines through a ring-closing metathesis-enyne metathesis tandem reaction.
